Bat Species Identification Service from droppings

A robust method has been established to allow for fast turn around and competitive pricing of bat dropping analyses. We’re located in Derby, UK so sending samples to us is easy and we are well versed in the identification of UK bats.

The processing of bat droppings is as follows:

  1. Sample Collection

Collection kits can be ordered here. They contain everything you need to collect and preserve samples between collection and analysis. The price includes return shipping so it’s a popular, cost effective choice. Order by 1PM to receive them next-day. Kits have a 2 year expiry so you can stock up and use as needed.

Alternatively, if you prefer, please collect samples using your own equipment and send them to our location in Morley. If you choose this option, please ensure this form is completed and included with the samples.)

  1. Sample receipt to the laboratory and booking

Upon receipt in our laboratory, samples are booked into our automated system and refrigerated to limit sample degradation. You will receive a confirmation email that they have been received and the analysis deadline date.

  1. DNA Extraction

A commercial DNA extraction kit is used to extract and concentrate the DNA present in the sample. Negative extraction controls are included to check for contamination.

  1. Amplification

Short fragments of a mitochondrial gene region are amplified with polymerase chain reaction (PCR). Special primers are used that amplify bat DNA. If a sample is very degraded, a restorative enzyme treatment is used.

  1. Purification and Genetic Sequencing

PCR products are run on an electrophoresis gel to confirm expected product size, and then purified using a commercial clean-up kit. The DNA concentration is calculated, normalized, and then Sanger sequenced.)

  1. Interpretation

The sequence is matched against a database of known species, and we are able to assign the origin species of the bat dropping.

  1. Verification and Reporting

The results are entered into a report, checked by a supervisor, and send to you via email. Be wary that sometimes results are sent to SPAM folders. A sample report can be found here.

Alcathoe bat- Myotis alcathoe

Barbastelle- Barbastella barbastellus

Bechstein’s bat- Myotis bechsteinii

Brandt’s bat- Myotis brandtii

Brown long-eared bat- Plecotus auritus

Common pipistrelle- Pipistrellus pipistrellus

Daubenton’s bat- Myotis daubentonii

Greater horseshoe bat- Rhinolophus ferrumequinum

Grey long-eared bat- Plecotus austriacus

Leisler’s bat- Nyctalus leisleri

Lesser horseshoe bat- Rhinolophus hipposideros

Nathusius’ pipistrelle- Pipistrellus nathusii

Natterer’s bat- Myotis nattereri

Noctule- Nyctalus noctula

Serotine- Eptesicus serotinus

Soprano pipistrelle- Pipistrellus pygmaeus

Whiskered bat- Myotis mystacinus

Due to the robustness of the service, the analysis will also detect rarities and vagrants such as Greater mouse-eared bat; Myotis myotis, Kuhl’s pipistrelle;Pipistrellus kuhlii, parti-coloured bat;Vespertilio murinus. If the droppings are from another species such as a mouse or bird, the results may detect this though not guaranteed to as the chemistry is tailored to the bats above.

Development of the Bat Species Identification Service

Hundreds of dropping samples were collected directly from all 17-breeding species and analysed to verify this service. Additional trials were conducted to optimise collection conditions and DNA extraction protocols. The results strongly indicated the least amount of DNA degradation when stored in the DNA preservative upon collection. A blind trial was conducted and gave 39/39 correct results.
